AIDS scientist charged with fraud

I have long held that scientists, like many other professionals, should be subject to a sort of “product liability”, if they employ fraud, engage in some other misconduct or in some way fail to meet the standards to be reasonably expected.

If a scientist is to be considered “responsible” for his work then this must be mirrored by a corresponding “liability”. In my experience a lack of liability is always accompanied by the absence of responsibility.

The product that researchers and scientists produce is publications – mainly as papers published in scientific journals and as books. Scientific misconduct (whether plagiarism or faking data or inventing data or cherry picking data) leads occasionally to dismissals (but not always) and generally very little else. It seems to me that the concept of tort or “product liability” should be applicable to the work of scientists and researchers where their work is the result of faking data, fraud or other misconduct since it would be work that “had not been done in good faith”. Tort would apply because the ramifications of their misconduct would extend far beyond their employment contracts with their employers.

Now an AIDS scientist who faked his data is being charged with criminal offences.

Responding to a major case of research misconduct, federal prosecutors have taken the rare step of filing charges against a scientist after he admitted falsifying data that led to millions in grants and hopes of a breakthrough in AIDS vaccine research.

Investigators say former Iowa State University laboratory manager Dong-Pyou Han has confessed to spiking samples of rabbit blood with human antibodies to make an experimental HIV vaccine appear to have great promise. After years of work and millions in National Institutes of Health grants, another laboratory uncovered irregularities that suggested the results – once hailed as groundbreaking – were bogus. 

Han was indicted last week on four counts of making false statements, each of which carries up to five years in prison. He was set to be arraigned Tuesday in Des Moines, but he didn’t show up due to an apparent paperwork mix-up. A prosecutor said Han will be given another chance to appear next week. …….

Experts said the fraud was extraordinary and that charges are rarely brought in such cases. The National Institutes of Health said it’s reviewing what impact the case has had on the research it funds.

…… Oransky, a journalist who also has a medical degree, said there have been only a handful of similar prosecutions in the last 30 years.

He said Han’s case was “particularly brazen” and noted that charges are rarely brought because the U.S. Office of Research Integrity, which investigates misconduct, doesn’t have prosecution authority, and most cases involve smaller amounts of money. …… 

According to the indictment, Han’s misconduct caused colleagues to make false statements in a federal grant application and progress reports to NIH. The NIH paid out $5 million under that grant as of earlier this month. Iowa State has agreed to pay back NIH nearly $500,000 for the cost of Han’s salary.

Han’s misconduct dates to when he worked at Case Western Reserve University in Cleveland under Michael Cho, who was leading a team testing an experimental HIV vaccine on rabbits. Starting in 2008, Cho’s team received initial NIH funding for the work. Cho reported soon that his vaccine was causing rabbits to develop antibodies to HIV, which left NIH officials “flabbergasted,” according to a criminal complaint against Han. Cho’s team sent blood samples in 2009 to Duke University researchers, who verified the apparent positive impact on the vaccinated rabbits. The confirmation was seen as “a major breakthrough in HIV/AIDS vaccine research,” according to the complaint.

Iowa State recruited Cho in 2009, and with his team – including Han – he soon received a five-year NIH grant to continue the research. The team kept reporting progress. But in January 2013, a team at Harvard University found the promising results had been achieved with rabbit blood spiked with human antibodies.

An investigation by Iowa State pinpointed Han, after he was caught sending more spiked samples to Duke University. In a Sept. 30, 2013 confession letter, Han said he started the fraud in 2009 “because he wanted (results) to look better” and that he acted alone.

Individual researchers are unlikely to have the means to make restitution for all the financial waste they may have caused by their misconduct. Universities and Institutions have  some possibility of being forced to repay grants obtained by fraud but are rarely asked to do so. Careers of other researchers could also have been compromised.

It should still be possible for someone damaged by scientific misconduct to make a civil case for damages even if criminal charges are not brought.  But what that needs is that the output be considered “product” and that the scientist and his institution have then some “product liability”. That implies a duty of good faith and of application of some reasonable level of competence. Misconduct and even gross negligence on the part of the institution or the scientist could then give rise to a claim for damages. Even the journals, their editors and reviewers  ought to have some responsibility and potential liability.

If every published paper carried some product liability, the rush to publish nonsense and lies may reduce even if the publications industry would not be pleased. But it would improve the quality of publications no end.

Marc Hauser actively manipulated data

Marc Hauser – and his supporters – have generally maintained that his misconduct was – at worst – negligence and certainly inadvertent. But the Boston Globe today reports on an internal Harvard report (obtained under FoI) which details wrongdoings rather more deliberate and sinister than Hauser and his friends have ever acknowledged or admitted.

The report is fairly damning.

But a copy of an internal Harvard report released to the Globe under the Freedom of Information Act now paints a vivid picture of what actually happened in the Hauser lab and suggests it was not mere negligence that led to the problems. 

The 85-page report details instances in which Hauser changed data so that it would show a desired effect. It shows that he more than once rebuffed or downplayed questions and concerns from people in his laboratory about how a result was obtained. The report also describes “a disturbing pattern of misrepresentation of results and shading of truth” and a “reckless disregard for basic scientific standards.”

A three-member Harvard committee reviewed 40 internal and external hard drives, interviewed 10 people, and examined original video and paper files that led them to conclude that Hauser had manipulated and falsified data.

Their report was sent to the federal Office of Research Integrity in 2010, but it was not released to the Globe by the agency until this week. ……… Much has been redacted from the report, including the identities of those who did the painstaking investigation and those who brought the problems to light.

Hauser, reached by phone Thursday, said he is focused on his work with at-risk youth on Cape Cod and declined to comment on the report.

The manipulation reported dates back at least to 2002 where he reported (presumably manufactured) data on a videotape of monkey responses which did not exist. In 2005 he altered data to make what was statistically insignificant become significant. Also in 2005, he discarded data after it had been found by a subordinate to have been inconsistent (presumably manipulated). Later, he tried to claim his mail ordering the discarding of the data as evidence of his innocence:

“These may not be the words of someone trying to alter data, but they could certainly be the words of someone who had previously altered data: having been confronted with a red highlighted spreadsheet showing previous alterations, it made more sense to proclaim disappointment about ‘errors’ and suggest recoding everything than, for example, sitting down to compare data sets to see how the ‘errors’ occurred,”

In 2007,

 a member of the laboratory wanted to recode an experiment involving rhesus monkey behavior, due to “inconsistencies” in the coding. “I am getting a bit pissed here. There were no inconsistencies!” Hauser responded, explaining how an analysis was done. 

Later that day, the person resigned from the lab. 

Conclusion that Förster manipulated data is “unavoidable”

Retraction Watch has now obtained and translated the report of the investigation by the Dutch National Board for Scientific Integrity (LOWI) into the suspicions about Jens Förster’s research. The conclusions are unavoidable that data manipulation must have taken place and could not have been the result of “sloppy science”.

Here are some of the highlights from the document, which we’ve had translated by a Dutch speaker:

“According to the LOWI, the conclusion that manipulation of the research data has taken place is unavoidable […] intervention must have taken place in the presentation of the results of the … experiment”

“The analyses of the expert … did add to [the Complainant’s report] that these linear patterns were specific for the primary analyses in the … article and did not show in subsets of the data, such as gender groups. [The expert stated that] only goal-oriented intervention in the entire dataset could have led this result.”

“There is an “absence of any form of accountability of the raw data files, as these were collected with questionnaires, and [there is an] absence of a convincing account by the Accused of the way in which the data of the experiments in the previous period were collected and processed.”

“[T]he assistants were not aware of the goal and hypotheses of the experiments [and] the LOWI excludes the possibility that the many research assistants, who were involved in the data collection in these experiments, could have created such exceptional statistical relations.”

What is particularly intriguing is the method of statistical investigation that was applied. Suspicions were not only because the data showed a remarkable linearity but that sub-sets of the data did not. The first suggests confirmation bias (cherry picking) but the second brings data manipulation into play. Non-linearity in sub-sets of data cannot just neatly cancel themselves out giving – fortuitously for the hypothesis – a linearity in the complete data set. The investigation methods are of more value than the Förster paper to be retracted.

I have an aversion to “science” based on questionnaires and “primed” subjects. They are almost as bad as the social psychology studies carried out based on Facebook or Twitter responses. They give results which can rarely be replicated. (I have an inherent suspicion of questionnaires due to my own “nasty” habit of “messing around” with my responses to questionnaires – especially when I am bored or if the questionnaire is a marketing or a political study).

Of course priming works— it couldn’t not work. But the lack of control over the information contained in social priming experiments guarantees unreliable outcomes for specific examples.  ..  

This gets worse because social priming studies are typically between-subject designs, and (shock!) different people are even more different from each other than the same people at different times! 

Then there’s also the issue of whether the social primes used across replications are, in fact, the same. It is currently impossible to be sure, because there is no strong theory of what the information is for these primes. In more straight forward perceptual priming (see below) if I present the same stimulus twice I know I’ve presented the same stimulus twice. But the meaning of social information depends not only on what the stimulus is but also who’s giving it and their relationship to the person receiving it, not to mention the state that person is in.

… In social priming, therefore, replicating the design and the stimuli doesn’t actually mean you’ve run the same study. The people are different and there’s just no way to make sure they are all experiencing the same social stimulus, the same information

And results from such studies, if they cannot be replicated, and even if they are the honest results of the study, have no applicability to anything wider than that study.

One would have thought the credentials of social psychology as a science – after Diedrik Staple, Dirk Smeesters and Mark Hauser – could not fall much lower. But data manipulation in social psychology would seem to be a bottomless pit.

Another case of data manipulation by social psychologists has erupted at the University of Amsterdam. This time by Jens Förster professor of social psychology at the University of Amsterdam and his colleague Markus Denzler. 

Retraction Watch: 

The University of Amsterdam has called for the retraction of a 2011 paper by two psychology researchers after a school investigation concluded that the article contained bogus data, the Dutch press are reporting.

The paper, “Sense Creative! The Impact of Global and Local Vision, Hearing, Touching, Tasting and Smelling on Creative and Analytic Thought,” was written by Jens Förster and Markus Denzler  and published in Social Psychological & Personality Science. ….

Jens Förster is no lightweight apparently. He is supposed to have research interests in the principles of motivation. Throughout my own career the practice of motivation in the workplace has been a special interest and I have read some of his papers. Now I feel let down. I have a theory that one of the primary motivators of social psychologists in academia is a narcissistic urge for media attention. No shortage of ego. And I note that as part of his webpage detailing his academic accomplishments he also feels it necessary to highlight his TV appearances!!!!

Rolf Degen: I hope that this will not escalate, as this could get ugly for the field of psychology. Jens Förster, a German, is a bigger name than Stapel ever was. He was repeatedly portrayed in the German media, not the least because of his second calling as a singer and a cabaret artist, and he has published an enormous amount of books, studies and review papers, all high quality stuff

This revelation occurs at a bad time for Förster, write the Dutch media. He is supposed to work as “Humboldt professor starting from June 1, and he was awarded five million Euros to do research at a German university the next five years. He is also supposed to cooperate with Jürgen Margraf – who is the President of the “German Society for Psychology” and as such the highest ranking German psychologist.

More stem cell fakery as a quick way to publication and fame?

Another young researcher, Dr Haruko Obokata has apparently made sensational claims about her stem cell research, shot to fame as lead author in two papers published in Nature and is now in the dock for dodgy images and irreproducible results (perhaps faked).

WSJ: Her co-author, Teruhiko Wakayama of Yamanashi University in Japan, called Monday for the retraction of the findings, published in late January in a pair of papers in the journal Nature.

The papers drew international attention because they held out a safer, easier and more ethical technique for creating master stem cells. These cells, which can be turned into all other body tissues, promise one day to transform the treatment of various ailments, from heart disease to Alzheimer’s. 

But shortly after the papers appeared, Japan’s Riken Center for Developmental Biology, where the work took place, began to investigate alleged irregularities in images used in the papers. Separately, many labs said they couldn’t replicate the results.

A spokesman for Riken said Tuesday that the institution was considering a retraction and that the article’s authors were discussing what to do.

Dr. Wakayama said he has asked the lead author, Haruko Obokata, to retract the studies. “There is no more credibility when there are such crucial mistakes,” he said in an email to The Wall Street Journal.

Dr. Wakayama said he learned Sunday that an image used in Dr. Obokata’s 2011 doctoral thesis had also been used in the Nature papers. “It’s unlikely that it was a careless mistake since it’s from a different experiment from a different time,” he said.

Like several other researchers, Dr. Wakayama said he hasn’t yet been able to reproduce the results. “There is no value in it if the technique cannot be replicated,” he said. 

But another co-author of the papers, Charles Vacanti, a tissue engineer at Harvard Medical School and Brigham and Women’s Hospital in Boston, defended the work. “Some mistakes were made, but they don’t affect the conclusions,” he said in an interview Monday. “Based on the information I have, I see no reason why these papers should be retracted.”

Dr. Vacanti—whose early work some 15 years ago spurred the novel experiments—said he was surprised to hear that one of his co-authors asked for the retraction.

Dr. Vacanti said he had spoken to Dr. Obokata on Monday and that she also stood by the research. “It would be very sad to have such an important paper retracted as a result of peer pressure, when indeed the data and conclusions are honest and valid,” said Dr. Vacanti. …..

The papers created a stir because they reported a process by which mouse cells could be returned to an embryonic-like state simply by dipping them in a mild acid solution, creating what they called STAP cells, for stimulus-triggered acquisition of pluripotency. ….

There seems to be a hint of some “academic rivalry” here as well.

Nature told the WSJ that it was still investigating the matter. As Nature‘s news section reported last month, lead author

…biologist Haruko Obokata, who is based at the institution…shot to fame as the lead author of two papers12 published in Nature last month that demonstrated a way to reprogram mature mouse cells into an embryonic state by simply applying stress, such as exposure to acid conditions or physical pressure on cell membranes.

Stem cell research seems to have more than its fair share of dodgy papers – presumably because sensational results are easier to come by and very much easier to get published.

Copenhagen Zoo’s justifications for killing Marius if applied to humans

Copenhagen Zoo has been marshalling support on the scientific and the ethical plane to try and justify their decision to kill Marius the healthy giraffe. They protest too much and it is a revealing exercise to apply their argumentation to humans.

Their basic theme is that He died so that others could live

Capital punishment could be applied for all humans convicted of murder or  causing a fatal accident or whose genes are defective in any way so that others may live. In current Danish politics, the wrong skin colour in a human is indicative of defective genes.

Culling is for the greater good of the giraffes

The man who pulled the trigger, the zoo’s own veterinarian Mads Frost Bertelsen, says that a very positive situation lies behind the Zoo’s action. 

”Up until now, we have not had to cull the giraffes. But now we have reached the point where the population is doing so well that a giraffe like Marius could not be relocated. Then the best solution is to put him down,” says Mads Frost Bertelsen.

The vet explains that a central European coordinator keeps track of pedigrees, and which genes are represented by individual giraffes in European zoos. The coordinator estimated from these data that Marius’ genes were already well represented and recommended that Marius was killed to protect the population best suited to the gene pool.

But now we have reached the point where the human population is doing so well in so many countries. Many individuals cannot be relocated. From East Europe or Africa to Europe for example. Then the best solution is to put them down, especially if their genes are already well represented. Something like the policy China had. Enforced abortion for all children after the first. 

The right time for Marius to die

Marius was allowed to live for one and a half years, then that was it. At that age he can, according to Bertelsen, be described as a ‘teenager’. It was an age when his father had also started roughing him up.

“In the wild he would leave the herd. If he were lucky, he would meet and join up with other young male giraffes. If he were  unlucky, he would be killed by lions,” says Mads Frost Bertelsen, explaining that it was not unnatural for Marius to die young.

In fact, the young male giraffes are most at risk of being killed and eaten on the savannah, because they do not have the protection of the herd when they are looking for females to mate.

If contraception or abortion are not permitted then the individual can be allowed to live for a while and put down just before it reaches child-bearing age. Lions and other carnivores could soon develop a liking for human flesh.

How to lead a natural life in the zoo

The Copenhagen Zoo lets the animals breed because one of the biggest challenges of keeping animals in captivity is that they are bored. …… a great activity for the captive animals is to find a partner, nest, have offspring, feed an raise their offspring, and finally spend energy on throwing the kids out.

“The side effect is that we have a surplus of animals. It is in fact fortunate that we can use them as food. Instead of killing 20 goats or a cow, we can use the giraffe,” says Mads Frost Bertelsen. ….. 

“Our function is not to keep the individual animal alive, but to keep the species alive,” says the Jens Sigsgaard and continues:

“We have decided that even if an animal is over-represented in the gene pool, we will let it breed and have as normal a life as possible. We prefer to kill ‘surplus animals’ rather than send them to zoos we cannot approve.”

For defective humans or humans of low intelligence, breeding could be encouraged as an antidote to boredom. Surplus individuals produced by such breeding can always then be culled and used as food. They should be killed rather than being sent to countries unwilling to accept them or to countries which cannot be approved.

The adult animals breed – the young must die

Aalborg Zoo has several arguments for allowing animals to breed, even if it may result in too many babies. …. “The animals are allowed to breed because it is an important part of their natural behavior to have offspring and experience the process of taking care of the them. Looking after the young is one of the best and most natural ways to occupy animals in captivity, In the wild there comes a time when the baby is old enough to break away from the mother and maybe become part of another group. That is the time when we try to find another well-suited zoo for it. If that is not possible, the young animal must be put down,” ”says Jens Sigsgaard. 

The animals can also be adversely affected if they are not allowed to breed and have offspring. They may find it difficult ever to start breeding again. And if there are no kids in the flock, the younger animals will not get the experience of what it is like to care for babies. 

The humans with the defective genes are allowed to breed as part of their natural behaviour. But when any young individual is old enough to break away from the mother we can try and find a new location for the individual. If that is not possible then it must be put down.

It is not the killing of an animal that is the problem; it is allowing the individual to be bred with the intention of killing it (and where the feeding of the carcass to lions is only incidental). And there is a difference in the breeding of mice for the purpose of being fed to snakes.

Animals are kept captive and alive in zoos just for gawking at. Once upon a time we did that with human “freaks”. I would like to think that we are more “civilised” now where I take “civilised” to be elegance in behaviour. The behaviour of Copenhagen Zoo with Marius was particularly inelegant.

The fundamental issue is that Copenhagen Zoo – like all zoos – are places for human entertainment.  They fool themselves – and others – into thinking that they are performing a scientific or conservation function – but that is just twaddle. (That is also the fundamental flaw in the conservation of species in zoos where – instead of trying to get the species to adapt genetically – the zoos try to “freeze” the animals genetically in a frozen and artificially maintained habitat).

There is something lacking in the ethics of Copenhagen Zoo – and all zoos for that matter.

I hope visitors to Copenhagen Zoo dry up….

One wonders what the purpose of the Copenhagen Zoo is? First they breed them. Then if they don’t like them or find them surplus to requirements they kill them. They invite zoo visitors to the autopsy. Why not to the executions? A healthy, 18 month old giraffe bred by the Copenhagen Zoo was killed off for being surplus to requirements (genetically)! Why breed it in the first place? And apparently this is standard practice.

If it had been culled in the wild because of an excess population it might have felt different. And the zoo had been offered alternatives.

An online petition to save the life of a young giraffe at Copenhagen Zoo is currently accumulating close to 2,000 votes an hour. But all the votes are in vain because the unfortunate animal was destroyed this morning in accordance with the zoo’s policy on in-breeding. 

The zoo said it has taken the decision to kill the 18-month-old male giraffe Marius and feed him to some of his fellow animals at the zoo in order to keep the giraffe population “genetically sound”.

“Giraffes today breed very well, and when they do you have to choose and make sure the ones you keep are the ones with the best genes,” Bengt Holst, the scientific director at the zoo, explained to the BBC.

Between 20 and 30 animals are put down in a similar fashion every year, added Holst. According to Ekstra Bladet tabloid, this has included bears, tigers and zebras.

…. Marius was killed by a bolt gun instead of a lethal injection, which would have contaminated the meat.

While most of him will be fed to the carnivores at the zoo, part of his carcass will be used for scientific research. Visitors to the zoo on Sunday were invited to attend the autopsy.

Ottawa CitizenCopenhagen Zoo turned down offers from other zoos and 500,000 euros ($680,000) from a private individual to save the life of a healthy giraffe before killing and slaughtering it Sunday to follow inbreeding recommendations made by a European association.

The 2-year-old male giraffe, named Marius, was put down using a bolt pistol and its meat will be fed to carnivores at the zoo, spokesman Tobias Stenbaek Bro said. Visitors, including children, were invited to watch while the giraffe was dissected.

Marius’ plight triggered a wave of online protests and renewed debate about the conditions of zoo animals. Before the giraffe was killed, an online petition to save it had received more than 20,000 signatures.

Stenbaek Bro said the zoo, which now has seven giraffes left, was recommended to put down Marius by the European Association of Zoos and Aquaria because there were already a lot of giraffes with similar genes in the organization’s breeding program.

The Amsterdam-based EAZA has 347 members, including many large zoos in European capitals, and works to conserve global biodiversity and to achieve the highest standards of care and breeding for animals.

Bengt Holst is the scientific director of Copenhagen Zoo who has implemented this policy of the EAZA.

Zoos are of course just places of entertainment for humans and I don’t really think they perform any other useful function. And I don’t much believe that their much vaunted  “conservation” which consists of freezing an unsuccessful species in an artificial habitat does that species any service at all.

The Purdue University School of Nuclear Engineering is the unlikely location for a tangled, sordid tale which I cannot make much sense of.

Professor Rusi Taleyarkhan is either a somewhat naive victim of a nasty conspiracy or he is guilty of academic misconduct and has received his just deserts. But his primary anatgonist was Professor Lefteri Tsoukalas once the head of the School of Nuclear Engineering but who was forced to resign as head by Purdue. Purdue removed Taleyarkhan’s endowed professorship, reduced his salary, and limited his duties with students. There is a murky connection between a journalist Eugenie Reich and Tsoukalas while Reich was promoting her book about scientific misconduct and there was some form of cooperation between them and a number of others to accuse Taleyarkhan.

Once I got this far I gave up.

There is quite obviously a great deal of muck in the Purdue University School of Nuclear Engineering. The University is probably vacillating between support for the warring academics. The role of  the journalist is what adds to the possibility of a nasty conspiracy.

It seems too tawdry to waste much time on though, of course, some careers are being destroyed and someone is – or both are – indulging in defamation.

The New Energy Times has a whole series of articles on the subject. They seem to feel that Talayarkhan has been badly wronged. This article in TwoCircles also takes that position. Tsoukalas puts his position in a letter provided to the New York Times (in 2007).

Oh what a tangled web they weave. It’s all about low-energy, table-top fusion — so it is all probably a hurricane in a thimble. I cannot help observing that cold fusion and claims of misconduct generally seem to go hand-in-hand!

But minutes later, Professor Turney drove six more passengers into the field. The overloaded vehicle had no space to collect returning passengers.

But there are still unanswered questions as to who will pay for the expensive international rescue mission. The Aurora Australis had to suspend a resupply mission to Australia’s permanent base in the Antarctic, Casey Station, to take part in the rescue.

The Shokalskiy’s captain, Igor Kielev, did not respond to Fairfax Media’s emails.

Chris Turney and Chris Fogwill, the expedition leaders, also declined to comment on specific questions regarding events on December 23.

Nicky Phillips and Colin Cosier travelled on board the Aurora Australis as part of the Australian Antarctic Division’s media fellowship program.

Amazingly, Chris Turney gets an award for “contributing to the understanding natural phenomena”. I suppose it’s a case of rewarding the Fool Who Rushed In!!

Pfizer writes off $725 million – were they a victim of scientific fraud?

Whether it is just error or bad judgement or fraud we will never know. Perhaps all three.

NewsObserver: In 2008, Pfizer paid $725 million for the rights to a Russian cold medicine called Dimebon. The pharmaceutical giant thought the drug could help ameliorate the symptoms of Alzheimer’s. Several clinical trials showed the medicine had no more impact than a placebo. Pfizer has largely abandoned the project.

Earlier this week came the news that Pfizer have now written off the entire $725 million.

The last flickering hope that Medivation’s Dimebon could help Alzheimer’s disease patients has just been extinguished. The biotech announced this morning that a 12-month study of the drug failed to register significant improvements for patients, mirroring two shorter Phase III studies in which Dimebon failed to outperform a sugar pill. Pfizer took the opportunity to bow out of its partnership, writing off its $225 million upfront and $500 million milestone program for what proved to be another embarrassing pipeline failure.

In 2008, Dimebon looked like an odds-on success, with positive data from a Russian study and 10 years of sales experience to underscore its safety. But Medivation was shaken to the core when its first late-stage study ended in failure, with an additional pratfall for Huntington’s disease to cap the disaster.

In the end, Dimebon’s failure helped tarnish the reputation of Russian drug studies while raising severe doubts about Medivation.

It is not only Pfizer which has been forced to make costly write-offs. Not long ago  GlaxoSmithKline was forced to shut down Sirtris Pharmaceuticals which it had acquired in 2008 for $720 million:

Glaxo paid $720 million to acquire Sirtris in April 2008, to get ahold of technology that generated lots of breathless media coverage as a modern-day fountain of youth. The company sought to make drugs that act on sirtuins, a class of proteins that scientists believe play a role in aging, programmed cell death, and other key cell processes.

Even though the company is closing the Sirtris site, Stubbee says Glaxo remains confident in the drug candidates it got from that acquisition. ….

…. Sirtuins are known to be active when the body is in a calorie-restricted state, which scientists have shown contributes to longer lifespan. The idea at Sirtris was to make small-molecule chemical compounds that activated sirtuins as a way of fighting diseases that develop as people age—including Type 2 diabetes and cancer. ….. The research into the biological role of sirtuins, from Sirtris co-founder David Sinclair, has attracted its share of skeptics. Just last week, Sinclair, a researcher at Harvard Medical School, sought to buttress his early work with a new article in Science that says resveratrol and related compounds can activate sirtuins. One critic, quoted by the Boston Globe last week, said the role of one sirtuin called SIRT1 in aging, “is still as clear as mud.”

GSK is putting a brave face on all of this.

But for many medical and biotech researchers, the path to fame and fortune is by starting a start-up with some new compound or technique and by “leveraging the promise”. For pharma and biotechnology start-ups the objective is to be bought up by one of the majors for as exorbitant an amount as can be managed. And it seems that one way to inflate the value is by making preliminary data and trials show very optimistic results. Negative results never see the light of day and the positive aspects are exaggerated and at worst manipulated.

The ten-fold growth of retractions in medicine related fields since 1975 is mainly due to misconduct according to this report in Nature where “fraud or suspected fraud was responsible for 43% of the retractions”.

For Big Pharma this is simply a consequence of having “outsourced” part of their research. They can afford a few failures it could be thought. Of course the final cost is eventually borne by the consumers but some start-ups make a killing along the way.
